NSAIDs in the Treatmet of Pain

Abstract

|
A few classes of drugs have been utilized for the control of pain depending upon the degree or severity of pain. Nonsteroidal antiinflammatory drugs (NSAIDs), simple analgesics, opioids analgesics, and adjuvant drugs are commonly prescribed by physicians. Here, the characteristics and clinical guideline of NSAIDs use are reviewed. NSAIDs can be classified by their chemical class, half life, and preference to COX-I/II inhibition. exert clinical effects by their analgesic, antipyretic, and antiinflammatory actions. Most of NSAIDs belong to weak acid except nabumetone, and inhibit prostaglandin synthesis by means of inhibiting cyclooxygenase (COX). COX has two isoenzyme at least, and two isozymes have different time dependent activation, distribution, response to noxious stimuli. Most of the adverse reactions could be derived from inhibition of physiologic function of prostaglndins, however lately developed COX-II specific inhibitors could reduce side reactions including gastrointestinal toxicity even though they still have limitations in renal, cardiovascular, neurologic system.
